Scientific Column Selection with ACD/Column Selector

Mike McBrien

Abstract

The United States Pharmacopoeia in 1970 introduced their approach to classifying stationary phase materials for chromatography. This system has several drawbacks. The most important is probably that many materials with the same USP classification do not exhibit the same chromatographic behavior. The result is that there is no accepted system for substitution of equivalent columns. If a column vendor discontinues a column, or that column is out of stock, there is no way to find an alternate. In addition, there is no scientific way to find columns that will exhibit different responses when required, and certainly no way to predict how other columns will behave. In this presentation, a system for evaluating and matching chromatographic columns will be shown. ACD/Labs has developed the freeware application, ACD/Column Selector, in collaboration with AstraZeneca to apply this technique in a practical way. Equivalent columns can be retrieved from a knowledge base of popular columns. In addition, orthogonal columns can also be retrieved.
